The easiest way to accomplish this is in the first level of Infinite mode.
It seems like more enemies spawn in at the end of the level, so get the warp gate to appear by rescuing all the villagers then position yourself in a smallish, partially closed off area and go to work. The chain meter is to the left of your health, you have a reasonable amount of time between hits which is good and when you take a hit it doesn't reset. It's a good idea to try to start with close to full health because it will reset if you die.

The easiest and safest way is to find a small room. More enemies seem to spawn there. The first level has one off to the left and only those goblin things spawn. The park yourself in a corner and mash the A button. You'll never take damage.
This is also an easy way to get the 1000 gold and million points achievement as well. I ended up with a 2000+ combo which netted me 27,000,000 points.
