Challenge Mode will be available from the start but it's highly recommended you play through the story first so you can get an idea of how to defeat all of the bosses (and since you won't get game over if you fail). Challenge mode is exactly as it says, a challenge. A good one at that. In Challenge mode, you get 1 life. All of the bosses are taken up a notch and you will get random perks/abilities that you can use throughout the fights. Due to the randomness, you really want to make sure you're choosing your abilities wisely. The order the bosses appear in will also be random so you can't even really prep properly for "the next one". This will undoubtedly take a few tries to get and I wish I could offer more to help but due to the RNG nature of this mode, it's near impossible to prep for it. If you found a loadout of abilities in story mode that worked really well for you, try and choose only those abilities to carry with you. You can equip/unequip anything you've unlocked so don't worry about having to use them every single time. Again, it'll undoubtedly take a few tries to get this but once you've completed Challenge mode, the trophy will unlock.
