You can get this achievement easily during the credits at the end of the game, but if you would like to get it sooner you can also easily get it on the first level as shown in the video below:

It should only take you about a minute even without upgrades.

There is no need to worry about grinding this one out at all. There are far more than 10 astronauts you can kill during the end game credit sequence. They will count towards this and you don't need to save them during this sequence for any other achievements

You monster. You can do this on any level or any mix of levels. You just need to kill any 10 Astronauts as you play through the game. The Astronauts that appear in the Credits also count towards this trophy so if you don't want to take a risk during the actual Campaign, there are more than enough Astronauts in the Credits to get you this trophy, even more so since you'll have to play through the Credits twice (once after your initial run and once after you complete New Game +). As soon as you've murdered your 10th innocent Astronaut, the trophy will unlock.
