You have to complete stage Z5.0, destroying 2400 of the little colourful blobs along the way.
(EDIT: AudibleJam509 suggests that this is cumulative across multiple attempts, and that any difficulty will do)
I wasn't counting, but it won't hurt to let those enemies that lay trails of blobs to do their thing before you clean up after them. And of course, the giant wall of blobs at the end should get you most of them.
This won't unlock on Practice difficulty. I unlocked it on Bydo, but it might work on Normal.

FYI: I unlocked this on practice. It’s cumulatively. Did the stage three times. First practice, second bydo, third practice, then it unlocked.
