You can use two controllers for this. Pick a run play for the offense and just switch (B) and dive (A) on the defense.

Dumb question, does this have to be exactly 50 yards? I've done long pass plays that go over 50 but no achievement.

The 50-yard pass has to literally be a throw that goes 50 yards in the air. I recommend equipping a card to allow more throwing distance.
