This achievement is unmissable, you WILL get this achievement as long as you complete all mission objectives. There will be no need for second playthroughs, as there's more than enough mandatory ground targets, you'll end up bombing 2-3 hundred no problem. You should get this on "Transport Weapons" which is the second mission of Operation Belt, the fourth campaign.
The quickest way to finish the mission and rack up ground kills is to strafe the enemy along the main road with machine gun and rocket fire. There will be one or two ground vehicles and 10+ infantry in the factory town and 3 more half tracks at the end of the mission. You won't get them all on one go, so just turn around, get back on the line you were on, and repeat.
Edit: While you can grind this achievement, as KP ScuzzyBunny points out in the comments below, there's absolutely no reason to do so, as I originally stated, this achievement is unmissable, there's no way you'll not be able to get the 100 targets, the only thing that you have control over is how soon you get it. KP got it before me, I have a few friends that got it after, depends on how much you go after non-mandatory ground targets.

I actually got it prior to finishing the 3rd campaign, so definitely no reason to try to grind it!

You're absolutely right, I rushed a little to get this up, let me edit it so it has the whole scope of the campaign in mind.

Story related; this trophy cannot be missed – You will unlock this trophy by naturally playing through the single player campaign since many of the missions require you to destroy ground units. While the title of this trophy is Bomber Ace, it only requires you to destroy ground units. So if you have difficulty using bombs, using rockets or machine guns will still count towards the 100. I unlocked this trophy during the mission Bomber Support from the Operation Belt single player campaign.
