The trait Vaccinated is unlocked by having a pupil get sick from mosquitoes. One of the Minor Vagaries, random events that occur during gameplay, triggers an event where traveling across a bog or jungle causes sickness. Pick a pupil, path its travel across one of these while the event is active (a green cloud will show on the tile), and a green icon will appear confirming the pupil is sick.
