After you've killed a few zombies there will be a counter at the left of the screen. This is the Killing Spree counter. You might have noticed it has a timer on it. After you have killed 25 zombies the Killing Spree will be extended, meaning the timer will reset and you will need to kill 25 zombies again. Rinse and repeat until you get to 100.
If you're having trouble with it, do it on a Checkpoint Race so the AI will leave you alone. Choose Bedlam ( the little buggy-like car ) he has a machine gun ! Pick one of the loadouts that will give you double ammo and choose any track that you like best ( and preferably know where to find the machine gun powerup ). Now just drive around, and keep the "A" Button pressed down. You should stay close to the machine gun powerup so you can get it when your out of ammo. This achievement will unlock as soon as you get your fourth Killing Spree extension.

This is also really easy to pick up on the Massacre Madness challenge. Grab the drunk missiles at the top of the long road to the left and just keep driving around the map - I achieved a 350 killstreak just doing that.

For me, I got this while I'm doing the Endurance challenge (I use Rampage loadout in order for me to boost early); I grab a harpoon, drive straight, shoot repeatedly, boost, turn back, drive straight, shoot, grab harpoon, and repeat.
