This trophy is cumulative, that is, if you destroyed 499 cars in one playthrough (which I doubt )))) and then started the game again and blew up one car, you will immediately get a trophy.

I got this trophy on my 4th playthrough.

I would like to note that not only cars, but also explosive barrels are counted.

There are quite a few cars scattered throughout the levels; you can blow them up simply by shooting, or using the very first barrel with unlimited ammunition and holding down until the sight becomes bolder and shoot at the car. Towards the end the trophy is yours.
