Here is a very simple and easy way to get this achievement.
Don't worry about trying to get this achievement using one of the power-ups (although you can). Just play the game as normal and play all the Challenges you come across (which you need to do anyway for 2 other achievements).
You will eventually get a Challenge where you have to kill "X" number of enemies using the Medusa power. You will have the power for the whole 30 second Challenge and there are a ton of enemies.
Just hold down the "X" button and shoot enemies until they shatter. If you only turn them to stone without shattering them, it will not count.
Also, you do not need to have all five frozen before shattering them, as stated in other solutions. You just need to shatter 5 total.
