Here is a very simple and easy way to get this achievement.
Don't worry about trying to get this achievement using one of the power-ups (although this one is fairly easy to get using a power-up). Just play the game as normal and play all the Challenges you come across (which you need to do anyway for 2 other achievements).
You will eventually get a Challenge where you have to kill "X" number of enemies using the Power Punch power. You will have the power for the whole 30 second Challenge and there are a ton of enemies.
Just start mashing the "X" button and hitting enemies until you defeat five of them.
