Should I train these character in conquest mode before story mode because some of them are hard to use?

It is up to you, I didn't as you have to play through with them and the XP carries across characters in this so they will all level up each other. In all honesty I would drop the difficulty if you are having trouble as it doesn't harm any achievements or anything to play on Easy vs. Hard
