I personally had this achievement before completing everything else however a friend of mine did not.
For most battles I was crossing the map by horse (The best horse would be Red Hare), so what I would do is for the most part ride in to hordes of nameless troops and just press X killing most of them depending on the weapon/difficulty.
Anyway, if you are going for a thousand and at least try to stop by the groupings in between officer battles you are sure to get this before you are done with anything else. Also don't be afraid to stop by places with a big group and use your musou to obliterate them, yeah it makes defeating officers a little trickier but unless you are on Normal or higher officers shouldn't be that much of a problem.
A good level to grind out kills is the stage where each force is attacking you (Shu , Wu , Wei , Other , Jin) because they bring a TON of troops and you can get some easy skill points while you are hacking away at the poor grunts in front of you!

There is nothing special to do to unlock this achievement.
Just keep going slashing the ennemies, it will eventually pop.
For my part, i had it during the Conquest mode, after i had finish all the Story Mode and i wasn't aiming specifiacally this achievement (undestrand by that : i didn't kill all people in every map).
If you are targeting this achievement, you can finish the Wei Story Mode then replay The Yangping gate battle where all the ennemies respawn almost forever.
