For this achievement, you must finish every single stage in conquest mode. There are about 300-400 stages all told. This includes every legendary battle (the tiles with a face on them). This means if a legendary tile has three battles on it, they must all be completed. Basically, if the tile is gold, you are done with it, if it's silver, you have work to do.
How long it takes really depends on how you play. If you rush the commander/objective, you are probably looking at 20-30 hours, if you play passively, 40-50 is more likely.
Note that there appears to be a bug with this. There is a weapon you get for finishing all the conquest stages, but you will more than likely get it before you do. I'm guessing it counts the number of stages you've done and when it hits the number of total stages, it gives you the weapon, not realizing some of those battles you fought were repeated battles.
This doesn't seem to affect the achievement unlocking at the proper time, though.

Best charcter to use in this game mode.

Since every character can use just about any weapon, the character you choose doesn't matter all that much. Increase in character stats doesn't seem to matter much either, so it's really all about the weapons and the seals you use. I rock axe and the curved sword, personally. Just about anything with good range and a decent attack will work well. The halberd also works well.
