"Exceed 100 plays" means 'play at least 100 missions'.
When you are playing the 'story' or offline mode, choose a pilot and go to the 'reports' option. On that list of info is a counter for how many missions you've cleared. You dont actually have to clear 100 missions, however, I hit it at 99 clears because I failed a Relationship mission and had to retry it.
It also does not take your Online missions into account.
