As the description states, this achievement is for destroying 100 Anthills or spider nests. These come in the form of large brown mounds coming out of the ground almost like a volcano and you will see multiple ants or spiders crawling out from the top.

Just to clear things up, blowing up the hills with pesticide weapons does NOT count towards the achievement. So this means that you have to go up and plant the charge yourself to destroy the ant hill. To make this easier, you can run up and unload an assault rifle in to the hill and it will eventually close off giving you a brief window to plant the charge, though in most cases you can plant the charge easily without having to do this.

A quick warning, if you are playing with bots be careful because they will unload on you if you are planting the bomb and they want to close the hill first. So I would advise circling around to the opposite side of them to save yourself frustration.

If you are only wanting to get the achievement quick, you can load up ditch slap and plant on the hill that is in the beginning and advance to the next point with 3 more ant hills behind the wall you have to blow up. After blowing them up, then restart the stage. You won't earn credits this way but your hills destroyed and kills will all count in the stats.

What's the best level to farm ant hills *and* credits? Also, note that "Ditch Slap" is level 1-4.

I personally had way more ant hills than I needed, because I ran the first level. Using the homing missiles of the jet it was easy enough to let the game sit there and kill the final ant hill while I sat back and did something else. i can't recall if that weapon was DLC though.

You destroy anthills/nest by going up to it and pressing/holding to set a charge. You should have 100 destroyed by the end of your three story playthroughs. If not grind them out on either Chapter 1, Mission 1 where there are at least 2 anthills or Chapter 3, Mission 1 where there are 4+ anthills. Additionally, at the end of Mission 4 on Chapter 3 there are 4 anthills that can be destroyed.

Tips on destroying them:

Anthills/Spider Nests can be destroyed in phases. Essentially when you approach a nest it will have the appearance of a small mountain with a circular top. Ants/Spiders will continue to pour out of the hill/nest when it is like this. You'll want to continually shoot at the nest/hill to suppress the flow of ants/spiders. When it is completely suppressed it will have the appearance of a small mound and that circular top will go away. In this phase the hill/nest is still active but no where near as many enemies will be pouring from it. At this point you can run up to it and press/hold with much less hassle. Make sure not to take too long planting the charge, otherwise it will open back up to its original state and the ants/spiders will start pouring out of it again.

It is possible once you get the pesticide gun to kill destroy these anthills with one shot. Please note that you won't get credit for destroying them this way. You have to set the charge by pressing in order for it to count for the trophy.

Phase 1 - Not suppressed and ants will flow freely
Phase 2 - Suppressed, ants will not flow freely, ideal time to set charge
Phase 3 - Completely destroyed, Woot!

If you don't get this while going for all the other Campaign achievements, there are six in Chapter 1, Mission 4 which is rather short to do. Destroying these with the Pesticide weapons does not count, you need to manually arm the mines to blow them up.
