** Due to the game being a Rogue Lite I cannot give an exact location to get the achievement ** however I can give you the method I used to get the achievement.
Method :
The achievement is for killing 100 enemies by melee (X Button) in one run.I suggest using your upgrade points to have max health as you can take a lot of damage going for this. Start at the beginning Comic 1 (Chapter 1) and melee attack every enemy you see. You can have the achievement knocked out before you start comic 2 if you attack every enemy in every room.
