Kill 500 enemies any way possible. This achievement will come eventually from just playing the game and likely to unlock in just a few missions. Follow the in-game tutorial for building towers, etc., start the levels and take them out.
This is a cumulative counting. Replaying any stage or attempting Heroic or Iron challenges all count as enemy kills.
These other achievements are similar:
FIRST BLOOD ... (kill 1 enemy)
SLAYER ...............(kill 2,500 enemies)
TERMINATOR .. (kill 10,000 enemies)
