Each Underboss has three unique associates which are unlocked based on there current earn. You do not need to complete the lieutenant side missions, you only need to introduce yourself to them. You will need to keep each underboss alive until all associates are unlocked.
Underboss & Lieutenant
Cassandra > Emmanuel
Vito > Alma
Burke > Nicki
Associates must all be unlocked during One Playthrough.
Associates are:
Jackie DuVernay, Arms Dealer - Cassandra
Jennifer Click's Moran, Operator - Cassandra (Earn 100k)
Jean Baptiste, Gunsmith, Cassandra (Earn 60k)
Fiona Davidson, Police Dispatcher -Burke (Earn 140k)
Hank McGahee, Vehicle Delivery - Burke
Terry Daly, Bomb Maker, Burke - Earn 100k
Bobby Navarro, Hit Squad - Vito - Earn 30k
Betty Johnson, Consigliere - Vito
Gianni Bruno, Mob Doctor, - Vito- Earn 60k
You will need to keep each Underboss alive long enough to reach the required earn to unlock there unique associates. For example if you reach Burkes Earn and unlock all three of his associates but he dies before you have unlocked Vito's/Cassandras associates then you will lose the achievement as Burkes Associates will be unavailable after his death. As long as Cassandra's Earn is at least 100k, Burkes Earn is 140k & Vitos Earn is 60k then the achievement will pop.

I only have 100k for Burke and it unlocked.

There are 6 associates, 2 for each Underboss (Cassandra, Vito, Burke) that will be unlocked as you conquer their respective starting districts. Associates provide you with perks such as a Mobile Arms Dealer, Vehicle Delivery, and Consigliere, in addition to upgrades that are unlocked as their respective underboss’ earning power increases. This will come naturally on your "Flip" playthrough by spreading the districts between your Underbosses and increasing their Earn.
