About 10 minutes into the game (if you don't skip the cut scenes) you be given a gauntlet and a quick tutorial on how to absorb souls.
Shortly after this you will come across 4 or 5 enemies attacking a wooden fort. Slice them up, when they're dead they'll leave behind some red floating orbs. As the tutorial states, hold down
to absorb them.

This will come naturally, but just incase, once you see the cutscene where the Ogre Clan give you the Oni Gauntlet, kill an enemy then press and Hold O to absord the souls of the enemies.

In Onimusha, you use Samanosuke's gauntlet to absorb the souls of demons you defeat pressing and holding 
. Just after the first encounter, you will receive the gauntlet. After this, just beat and absorb the souls of this first set of demons - then the trophy will pop.
