The achievement is called Professional Exterminator and requiers to kill the Gargantua on Hard in 2 minutes 30 seconds or less.
The Gargantua is the spider-like boss at the end of Scene 6. Its weak point is the mouth whenever it opens up to shoot at you, though you still can do somewhat minimal damage when its mouth isn't open. A way to make this fight slightly easier is to play the first parts of the chapter on Easy while conserving ammo, then quit to the title screen when this fight starts. Choose to continue from where you're from and you can change the difficulty to Hard.
