This will come naturally, and quite quickly in your playthrough of the games required 4 levels.
A "mini-region" just means a small portion of the map where you see the green flickers, the same as shown in the tutorial. Your radiation counter will go up when you are in the presence of radiation.
I got this by clearing the big areas of trash that populate many of the levels, as I wanted to farm for radiation coins to make this game trivial, but in theory any small area of radiation counts towards this. Just keep an eye out for clearings that have that green sparkle.
