This achievement is for the police chase part of the Weird Science mission, it begins at the part when Jimmy picks you up with his car.
You get infinite RPG shots and there is probably 100 or so total targets during the chase so you get plenty of opportunity. Don’t forget you can lock onto tougher to aim at military/police vehicles with
Helicopters, police cars, military suvs all should count towards the 45 needed.
Worst case scenario if you do mess up, try to make a save before you start the mission, just in case. This way you could always just try again if you miss it.

Adding to this: the far left Lockdown at the first roadblock can be avoided destroying (even with RPG) while killing everyone there. From there they keep sending helicopters(they count) and land vehicles at you. Easily completed that way quickly. Once completed or at the least getting to a satisfied amount of vehicles destroyed its guaranteed to get this.

Right after Technically Legal, Jimmy will come by to pick you up. Once you're in his car he will start to chase The Johnny Brute and whilst you're chasing Johnny, the Police and Swat will be chasing you. I did this with the infinite Rocket Launcher and it was still a little difficult. There are only about 60 police vehicles you can destroy so try to get it out of the way as soon as possible!
The trophy will pop once all 45 vehicles are destroyed.
