Unless you're charging through the game on a speed run you should get this easily through normal play. It popped for me towards the end of the Deadpool level so you should have no problem obtaining it before the end of the game, even if you're not taking down everything in sight.

I got this just by playing through the story. There are, however, a few levels that stand out, if you're in a rush.
The Unlimited level with Electro has a fair number of enemies, the majority of which are pretty weak.
The Unlimited level with Carnage has a massive amount of enemies, some weak and some that might take a minute or so.
Some people favor the Unlimited level with Deadpool, but I find it to be too much of a strain, and more complex than the previously said levels

You'll get this just by playing the game and trying to platinum it. All the enemies you fight and kill will all save. No need to keep trying to kill them, it will come naturally.
