I found this easiest to do in "The Village". The best way to boost this is to go to the upper ledge area on the main land, and go to the rock pile in the middle of this upper area. Now take the rocks, and aim in at the mound that the electro-flies are flying around, and a crawling bug will emerge. Hit this bug with a rock 3-4 times and it will die. Rinse and repeat 25 times, and the achievement is yours.
If you can correctly aim at the base of the mound, you can actually just continually keep throwing rocks in the same place, and unlock the achievement after 3 minutes.

This will come as you play the game normally as you will have to kill more than that just to get through everything. If for some strange reason you do not get this by the end of the game, just reload any chapter and find a firefly nest where they will respawn from after you kill them.
Note: Lightning Bugs are the ones that look like cockroaches that shoot electricity (There are fireflies that use electricity, those are NOT Lightning Bugs).
