The text for this achievement is misleading - this requires 50 cumulative victories in 1vs1 Mode, and can be done fairly easily if you've grinded several characters after accomplishing the 3vs3 Mode achievements beforehand.
There is also an issue where this achievement will not pop upon completion. I had to file a ticket with JoyCity to get them to unlock the achievement after hitting the 50 victories on my User Stats (found under Management). They requested my XBox ID, screenshots of my User Stats page, and the Achievement's tracking status.
The best way to secure the wins is to play on the now-default Full Control Mode, where you can control all characters. On defense, pressing in the
will switch you across the characters. Sometimes it's easier to control your big (C/PF) while the opponent is playing a small shooter (PG/SG) is trying to shake off their defender. Once the opposing player stops, that's when it's best to click onto their defender to avoid the AI from making a mistake that will open them up for a shot. Sometimes it's best to allow the AI to do its job, as sometimes it can make a critical block on a poor shot attempt or get a steal. On offense, switching is as simple as passing the ball to the character you want to control.
If Full Control Mode doesn't work out for you - for various reasons, such as the being sensitive and switching you when you're just trying to move the 
- then the key to winning through Single Control Mode is picking the character you feel most comfortable, calling for the ball near the start of every offensive possession, and just looking to make simple moves to get past the defender and get an open shot near or under the basket. A lot of the time, the easiest bucket to get will be a layup after blowing past your defender. Trying to fire off 3 point shots will be harder if you're F2P and don't have a ton of shooting Platinum cards, so your best bet will be to just look for ways to get the layup.

Thanks so much for mentioning this. I'm at 65 wins and already have two achievments glitched (50 MVPs and 50 substitutions). Where did you go to file a ticket and how quick was the process?

I recommend waiting until after you've finished 3 on 3 mode to even go for this unless it's driving you mad not having it sooner. The goal here is to pick your character and 2 AI characters to play against another team online. By the end of 3 on 3 mode, you will have (hopefully) 3 players at different positions leveled up. This will help out greatly when playing against others, as a 4-star AI player on your team is almost as good (and sometimes, better) as having a live player on your team. Win 50 matches and this is yours.
