I would suggest getting the $25 Million online winnings first, so you only have to get $25 million more, off or online.
There is a easy method to reach your $50 million winnings, which works in both online and offline modes. Play a game of Quickplay or Homeroom, and answer 6 questions correct to make it to the $50000 mark, then on the next question, DROP OUT. When you reach the winnings screen it will show $500000 instead of $50K, much easier and faster, as you don't have to worry about messing up on tougher difficulty questions.

You need to select the next kid & question and then drop out during the question.

The game does not give me the option of dropping out after six questions or at 50 000 $ .

This achievement requires you to win $50 Million in total cumulative winnings over all your games. This includes online as well as offline games. The fastest method is similar to the method I posted in Superintendent except that you don't need a boosting partner. Although it is much easier if you have a second controller so that you don't have to complete against the CPU.
My method:
Three And Out only penalizes you if you get the question wrong, but does nothing if you don't answer. Therefore wait until you see a question you definitely know the answer to and then answer it. You will have to answer the final question, and it is best to have a computer and Wikipedia, Google Maps, and a dictionary website.
Alternative Method: (Thanks Fluster Zero)
There is a glitch in homeroom mode when you get to $50,000 winnings, drop out and the game will award you with $500,000. This happens every time you do it. You would have to do this 50 times if you already have the $25mill achievement from online.
Try them both to see which way works best for you.
You also have lifelines up to that point which work %90 of the time and playing on easy seems to increase your odds to not failing if you get one wrong.
