From what I noticed in the Atomic Drill and YouTube and the challenge itself: Most of the house, as well as items, and family members locations are all randomized.

Info from Atomic Drill and challenge itself: 1 slot items: Cans of Soup (3 needed), Bottles of Water (3 needed), and the Map. 2 slot items/family members: Dolores, and Timmy. 3 slot items/family members: Suitcase, and Mary Jane.

Info from the challenge itself and YouTube: Other than getting to the bunker before the time runs out with the last items in hand, anything else is not necessary to pass, though some objects in the house and optional items may or may not get in the way, it also says that challenge mode disables most achievements, I wonder which ones it disables.

The Atomic Drill or Scavenge modes don't tell you what items are needed to pass, instead the challenge tells you, but both of those modes can be really useful for figuring out what items take up either 1, 2, or 3 slots, without compromising the challenge itself. I used the Atomic Drill to test what items take 1, 2, or 3 slots.
