How to unlock the Church Sanctuary achievement in Age of Empires II: Definitive Edition - Definitive Guide
One huge tip I can share for this one is to not destroy the castle at Tarsos until the end. The villagers count both in the early game when you are defending as well as the late game if you open them up as an ally.
If you destroy everything else and save the Byzantine troops around the city for last, there will be no chance of you potentially losing a villager.
In the beginning of the mission you are looking at 20 villagers that need to survive. I would suggest sending them en masse to the castle. You then just need to keep the trebs off the town center long enough to make it through the attack. When you flee the city, before losing control of the buildings, make sure you have 20 vills idle, otherwise reset.
IF you play with Tarsos joins on your team (destroying the castle that is in the city), you have no idea how many vills he had and if he loses any in the skirmishes with the Byzantines. This is unwise in any circumstance because even one lost villager later means the end of your achievement. Do yourself a favor, clean out the rest of the map, then come and sweep up the Byzantines in Tarsos.

It finally popped on my third try.
It seems a lot of people won't get it, if all the villagers are garrisoned when you lose control of the base.
Had to ungarrison them before the timer counted down.
Took the north base, and allied with Green.
Build Dromons, since you can take out Blue with only them from the water.
Just go for he's Castle and towers.
While you are allied with Green, build a castle and stables close their base, so you are ready to take them out after.
If you sent units inside the base, they will cancel the alliance.

Seems like the villagers garrisoned in the castle will die once the "reinforcement" timer runs out. Garrisoning them in the town centre works but you won't fit all 20. Better to put them in the castle and then ungarrison them when the timer is about to run out.
