The basic tips to give you are very much similar to Endless Mode, as your job is to survive while doing the missions. If you solely go for the missions you may find yourself with a full board:
1. Play with care and take your time if you have to.
2. Remember to use the Y button to use your items. Plenty of times I forgot it existed..the best times to use it are probably all at once when your board gets filled up.
3. When it comes to the couplers mission, I made sure to leave a coupler wide open that could easily be connected just for the purpose of that mission, but you should still try to connect as many couplers as possible otherwise they will make your life miserable.
4. Remember that pieces can be connected diagonally as well!
5. The next piece preview can help you decide on what you want to do next.
