The Soul Serpent can be found in the Swamp biome (Murkmire). It's a random encounter so just keep searching the swamp biome until he appears.
He's immune to damage until all four flowers are destroyed (one in each corner of the arena). You'll then get a small window to deal damage before the flowers regrow again. Rinse and repeat until he dies.
