This is best achieved during a replay of the chapter on Normal mode, so you can slide-kill any of the guards in your way. The time limit is rather generous, and resetting the checkpoint also resets the clock to its time when you originally triggered the checkpoint.
The only tricky part is near the middle, where there will be an elephant very close to you at all times. I recommend jumping during that stretch of the stage.
There is a boss fight at the end of the stage, so if you haven't made it to the final area between 4:30-5:00, consider restarting the stage for a faster time.

This is a speedrun sequence involving a boss fight at the end. Though not madatory, if you want to get the best rating for this sequence, you need to complete it under 6 minutes.
The speedrunning part is not too difficult, as killing enemies is allowed without voiding "Not Your Usual Assassin". Ideally, you should complete it under 4 minutes 30 seconds, so as to save as much time as possible for the boss fight, which can take longer than usual depending on your fighting skills.
