You can unlock this as early as DNA Sequence 1. For this achievement, you have to run continuously for 100 meters; it is not cumulative. All you have to do is hold down 
and 
to sprint without stopping or running into someone. This is easier to do around The Duomo in Central Florence where the streets are wider. Just run until the achievement unlocks. This takes about 15 seconds.

button to sprint. Run down a long street until the trophy pops up.
