With the latest patch, you now just have to reach level 200 and you will be given a mastery point to spend. If you are already higher than 200 then just boot the game after patching and go into the skills page to get the point. Spend it for the trophy

Spending your first Mastery point first requires that you reach power level 400, the max level in the game. This may seem daunting at first, but if you're going for all other achievements, you will reach level 400 far before you finish everything else in the game. Once you reach level 400, every time you level up after that you'll receive a Mastery point that you can use in your skill tree. Using your first one unlocks this achievement.
 
You gain XP to level up from completing quests, killing enemies, collecting "collectibles" all around the map (i.e. collecting wealth, and completing mysteries and artifacts on the map), discovering new locations, and syncing viewpoints. You will automatically get 2 skill points for each drengr and legendary animal you defeat, and you will automatically get one skill point each time you complete a standing stone puzzle, a cairn challenge, and an animus anomaly. The vast majority of your XP will come from completing story quests and World Events; you'll only get a small amount for artifacts and wealth collected. But as I said, you'll earn enough XP to reach level 400 well before you finish all of the story quests and complete all of the items in every region. To put it in perspective, by the time you finish all other achievements you'll have leveled up to the equivalent of level 450 or so (giving you, in that case, 50 mastery points to spend).

After reaching the maximum level of strength/power in the game (400), instead of points for learning skills, mastery points will be awarded - after the first one spent, the trophy will drop.

As of 11/11/2021, mastery points start to drop after power level 466, that is, after fully pumping the skill tree. You can easily make this trophy if you complete all additional events (condition, secrets, artifacts) until the end of the storyline.

As of 11/29/2021, the first mastery point drops after power level 469 .
These are all the story missions, half of England is cleared and the DLC storylines are completed.

As of 03/08/22 with all DLCs downloaded, the first master point appears after level 535

Carefully examine the skill tree. The game is updated periodically, and the tree is updated accordingly, new skills are added, and if they are not all open (not all skill points are spent), then mastery points will not be given.

After the release of the DLC Wrath of the Druids, you get your first Mastery point at level 428.

For those who don't have DLC.
At level 6 of the Yanli settlement, you will start selling a scroll of knowledge, which gives 1 skill point. The scroll of knowledge is updated when you gain a level OR after 3 hours spent in the game. Therefore, you should quickly pump up your settlements and take the scroll of knowledge when you level up, so as not to choke after completing everything.
If, like me, you've already completed everything, I recommend completing Red's quests (they're updated daily) and farming the city of Warheim (bottom left). There are a lot of strong enemies there, and stealth kills of them give 75 experience points (50 for a regular kill).
