You should already know how to do this from Chapter 1. There is one set of flowers in each of the rooms you go to to take photographs. The last 2 sets of flowers are in the mayor's room, one of which is on his desk.
In case you missed the instructions, here they are:
LEFT TRIGGER to lock on to a flower.
RIGHT TRIGGER to pollinate the flower.
RIGHT STICK to move to the next flower.
