Can confirm this works in chapter select. This was my last achievement so as a precaution I removed all my saves (offline and online) and uninstalled and reinstalled the game. Selected chapter 5 in chapter select and played through to the area, then finished the fight and popped the achievement as soon as Alice's dialogue started.
One thing I'd recommend is that, while it may seem obvious, don't move on to a different searcher until the one you hit is for sure dead. I noticed when I hit a lot of searchers they took more than one hit, even though it looked like they died. I really focused on this on my achievement run and I got it no problem, and my run wasn't my best in my opinion. Also for those curious, one run including loading screens is about 15 minutes, so while not ideal to have to go through the dialogue at the beginning again, it's not as bad as it may seem and it's significantly shorter than playing through the whole story again.

Impossible to unlock it. I did all the fights several times without dying with a lot of kill but nothing. does it pop right after the fight or later?

In all the videos none of them show the achievement pop so I was getting frustrated not knowing when. It happens right after Alice says "that is all of them."

So I unlocked this on 3-Dec-2022, so I can confirm this isn’t bugged and it is achievable.
That being said it can be tricky.
Firstly I used chapter select to start chapter 5, I’ve seen some guides say you have to do the entire campaign from the beginning but I used chapter select and that worked fine.
Next you have to sit through a 5 minute unskippable cutscene, really wish they’d add a skip button here.
Then grab the pipe and get yourself out of your room. Kill the 3 searchers on the way to the boat.
Get in the boat, sail down the ink river and obviously avoid the giant hand.
Once you make it to the dock you will encounter sammy, use the method of standing on that one barrel so you can hit him and he can’t hit you, after about 12 hits he will drop his mask, then walk up to him to trigger the next cutscene.
After this cutscene you will be joined by Alison and Tom. Now the method I used here was I watched YouTube videos so I knew where enemies would spawn and in what order, it might seem random but it’s not, it’s the same order every time.
Now I was under the impression that you had to kill more than Alison and Tom combined, not true. Tom tends to one-hit most things with his axe and Alison has a machete which also deals a lot of damage. So those two vs you, that’d be tough to get more kills than them. But the requirement for this trophy is to get 45 kills or more during this sequence without dying. So prioritise searchers and lost one’s, only go for miners if you have full health and if they’re distracted, and even then only go for one, because taking out more than one means you waste time when you could use that to get into position for the next round.
Watch your health, I found that if I took 2-3 hits I would back up to avoid dying, I would never go to the docks as this means Tom and Alison would take out all the enemies for me.
So watch health, memorise spawn points for enemies and the order in which they spawn, don’t die, take out at least 45 enemies (no, sammy and the 3 searchers and the beginning of the level do not count).
At the end Alison will say “I think that’s the last of them”, give it 30 seconds, if the trophy doesn’t pop by then that means you didn’t get enough kills and will have to start over.
This trophy can be very frustrating, so there’s no way to keep track of how many you’ve killed and you can be quickly overwhelmed. Just remember to start backing up a bit once you hit an enemy, means you still have range for a second attack with your axe, but that you can be out of range from their swipes.
