You will get this at the end of the game.
Once you have defeated both Big Sisters and Gathered the Big Sis suit for Eleanor you will be returned to your body and she will come rescue you. Upon that, this will unlock, you will also get a Plasmid that allows you to summon her (as a Big Sis) so she can help you in fights.
And it's story based, and therefore, cannot be missed.
