When you need to access a locked area, Elizabeth will pick the lock for you. There are many story-related locks that contribute to this, but if you want to get this without chapter reloading, you will need to unlock some of the optional locks.
Elizabeth will pick story-related locks with just her hairpin, so you don't consume lockpicks. However, optional doors consume between 1 and 5 lockpicks to pry open. In the early chapters of the campaign, picks will be scarce, so don't feel the need to pick every lock. Some collectibles are found behind locked doors, so keep some in your inventory at all times. Booker can carry up to 30 lockpicks, so use your inventory number to guide whether or not to unlock a door. If you unlock most of the doors you come across, and scavenge or buy as many lockpicks as possible, this achievement should come during natural gameplay.
You can also use reload chapter to get this. The best location would be the entrance to Soldier's Field. There's a locked door with a lockpick next to it on your left as you enter. Once Elizabeth has picked the lock, hit reload chapter and you will be back outside the door.
