In chapter 1, Nikolay will give you a personal quest to find his friend that went missing. You must speak with him in the home base before venturing out, otherwise you miss this. Just speak to all companions any time an exclamation is on their icon.
Once you have his quest, explore his personal side map to the end and once you fight the tree boss, this will unlock.
