Ok since the counter does not track your kills, the best place I found is on Level 1 after the plane mid-boss when you acquire the blue metal suit.
Play on normal and kill, kill, kill until you reach the mid-boss with the floating head attached to the hole. Just let him murder you and you should start at the beginning of that stage just before jumping in the suit. There is probably 50+ enemies here to kill in about a minute.

To make it little bit faster, there is hole just left of metal suit. Drop there 5 times and with your last life go killing. Either get killed very end on last enemies, or run to mid-boss and jump on it to die.

You can track your kills by selecting Achievements then Stats on the main menu.
