This one stumped me for a while, as I kept taking just a bit over 20 seconds using either the energy whip or grenade launcher. I was able to knock this out finally after using the charge laser.
As the elevator begins to move, fully charge the laser and nail the spider queen in the face with the full force blast as soon as the timer starts running, then use the normal shot from there on out. Repeat shooting up, pausing only to dodge the acid drops. When the purple cell things start dropping quickly take them out with the laser normal shot or melee, then resume shooting up at the head, dodging acid as needed.
With the laser I was able finish the queen off in about 16 seconds, and prevented her from even getting to the web attack.

You can do this in Boss Rush. I don’t know if it’s faster but you start with all weapons.

I got this my first time through the game on Easy difficulty. I didn't even realize this was an achievement until it popped after I killed the boss.
I had the weapon bot and just blasted her head with the base weapon until she died, avoiding a few projectiles in the process.

The boss is found in the second section of mission III. Make sure you have obtained the grenade launcher before getting here. Aim up and fire away while dodging the green acid spit, I was able to kill the spider in under 16 seconds.
