"Taking Flight" is the fourth mission in the game and has you getting Sanctuary III, ready for take off so follow Ellie's instructions and complete her missions. The trophy will pop, once you're in orbit and have spoken with Lilith.

Mouthpiece has the following attacks:
Soundwave Gun
Speaker Overload
Tink Spawn
Mouthpiece can be a pain to kill, because his Soundwave Gun and Speaker Overload attacks do a lot of damage if you get caught up in them. To avoid taking too much damage, stay behind him and move to a different area if you hear the speakers power up and see the cables on the floor turn pink. Mouthpiece is at his most vulnerable, when he's showing off and exposes his back to you so take advantage of this and empty your most powerful weapon into him. Once he's toast, pick up the Vault Key then return to Sanctuary III and give it to Lilith.
