If you're impatient you can go to score attack mode cave 4. There is a solid block of 78 diamonds in the upper left and diamonds are spaced fairly closely throughout the level so there should be no problem getting the remaining 22 before entering and/or after leaving this area.
Or in Arcade mode levels 75 and 98 are both essential one long line of diamonds, and the combo is pretty much a sure thing on either of them.

You have to get 100 diamonds without the counter resetting. There is a spot by your score showing you how many you got in a row. Select cave 75 or 98 in Arcade Mode to get 100+ chain combo easily. (Mahalo to Hawaiian for the information).
An alternate way of getting the 100 diamonds can be done if you know what you are doing. You can get it early by letting the amoeba take over 100 spaces then go and collect all gems in a row. (Thanks again to Turdboi and his Misses for the alternate suggestion).
