This achievement requires you to reach level 20 during a single run. You will level up from killing enemies throughout a run, and each time you level up you get presented with a choice of stat boosts. So, the more enemies you kill, the faster you'll level up.
My experience was that I surpassed level 20 on every successful run I had. I was typically level 21 or 22 in each run. And, that was without any items that increased XP gain; those runs, I reached level 25 or 26. You can grab one of those items if you see it for sale, but I found it unnecessary to reach level 20. You'll therefore likely reach level 20 during your first successful run.

Play on Danger 0 as well-rounded and make sure the settings on your game is 25% for enemy health, speed and damage. Start with an SMG and always get an SMG from the shop after every wave. Remember to combine 2 of the same weapons for higher damage. Then keep playing all the waves and keep collecting the green materials on the map that drop from the enemies and you'll see the green level bar fill up the more green materials you collect. If you don't get to level 20 by wave 20 just press endless when you're at the shop after wave 19.
