That being said it did not pop for me when I went through on my normal play through. It only popped when I went back through and beat the boss on very easy. No other guides say you need to do anything specific, though on my normal playthrough I died a bunch where on my very easy I beat the boss without dying.
If you don't get it the first time you can chapter select back and try again.

At the end of Act 4 - Chapter 2, you will fight the Hyper-Mutated Flytrap. This one of the only boss fights in the game but it isn’t too difficult of a fight. The goal is to shoot its mouth until the health bar is depleted, then shoot the orange spots to prevent it from regenerating. Use slide to stay mobile during the fight and avoid damage and plug away at it. I found the Flailgun to be strong during this fight, with the Peacemaker a good alternative for whittling down its health. Keep at it until it coughs up your friend to earn this trophy.
