Even on round 1 this isn't working for me.

This is much quicker if you run in the lab, as the Nova’s will spawn much more frequently. Though it will be much harder to run circles in there too.

Normal weapons seem to kill Nova 6 Crawlers and force them to explode. It doesn't matter what part of their body you kill them. I only found Winter's Howl and the Ray Gun at the moment that doesn't allow the Nova 6 crawlers to explode.

Should be noted knifing doesn't cause explosions either.

Nova Crawlers spawn after you turn on the power, mostly in the labs though later on in the rounds they spawn everywhere.
The Nova Crawlers will explode if you kill them with bullets. So don't use those. Instead, I suggest meleeing them. That's how I got the trophy. Buy the Bowie Knife in the labs and you can knife them with just 2 swipes up until at least round 25 (and at the most 28, I'm not too sure). And Instakill powerups are always useful.
I am told explosives also work so maybe spawn with the Welling and Pack-a-punch it if you desire.
You can do this on Casual difficulty. I suggest having 3 bots in your game. Yes they will steal some kills, but they will also distract the majority of the zombies, leaving you clear to focus on the Nova Crawlers. I strted attempt this pretty late, on like round 15 but I still didn't have trouble with normal zombies even on round 23.
Edit: there's a side quest that causes around 15-20 nova crawlers to spawn in, and it can be done on round one. Open the door across from the elevator, on the top floor, where a perk machine is. To the right on the wall of the Escargo gun wallbuy in the corner (left of the perk machine) is a door you can hold square on. You'll heard a quote. Continue holding square on the door every time the quote ends until you hear a quote from a twisted girl voice (Samantha). After she finishes, the nova crawlers will spawn in one by one. Just melee them.
Also, I noticed the Winter's Howl (the wonder weapon) doesn't make the nova crawlers explore, however I can't confirm if it works for the trophy.
