Stokers are the very large zombies carrying shovels that look like hardened magma. Not to be confused with the fire elemental zombies that detonate when you kill them.
After damaging the Stoker a certain amount, a weak spot will be revealed through a glowing yellowish-orange spot on either side of his chest or the top of his head. Shooting the glowing spot will cause him to drop to his knees and blow up from the weak spot.
Later rounds are recommended for this, as they spawn very frequently the higher you are. I did it on Round 32. There were about 6 or 7 across the entire round.

I would add that the Kraken won't assist with the achievement. I had Pack-a-Punch Green Kraken which would two shot them and it didn't pop the achievement.

To add to Racoonus' excellent guide on how to kill the stokers, a far more efficient way is to play on Rush mode as it is basically 1 continuous round. This way you can get this achievement without requiring high rounds

I can confirm that this no longer works playing Rush Mode. I have killed over 7 Stokers making very damn sure that my last shot was where the glowing spot was. No achievement after 25 minutes in the same game.

I got this by playing on casual with 1 bot and after round 20 you'll start to get 3 stokers every round. I used the zweihander and the mog 12 pack a punched
After spending far too long trying to get this achievement I think I found the best way of obtaining it.
Difficulty - Casual
Bots - 1
Loadout - MOG 12 (I had Cryofreeze ammo / yellow variant after PaPing)
This gun is also a wall buy if you are not high enough level to spawn with it.
Though it may sound counter intuitive, using the shotgun's bullet spread is what is going to be a lifesaver and help you hit those weak points without much mental effort. It's also going to help with making the stoker a quick kill, so you don't give the bot an opportunity to steal it, which happened to me more times than I'd like to admit while attempting this with other guns.
The bot is needed so that stokers spawn on earlier rounds. From everything I've seen, with 2 players (or 1 player and 1 bot) 3 stokers start to spawn at round 20. I got my achievement on round 19, so it's possible for them to spawn even earlier, but it seems rare as I never saw it happen outside of the one time.
As for perks, it's not important, but my loadout was Ethereal Razor, Death Perception, Winter's Wail, and Dying Wish.
Good luck, and I think the best advice I can give is don't get caught up trying to aim perfectly at the weak points. You're only going to give yourself a headache if you try hitting it flawlessly every time. Just grab your shotgun and shoot in the general area as best as you can.

Another thing to note: Stokers only spawn while other zombies are spawning and only 1 Stoker spawns at a time. Prioritize killing the Stoker so another may spawn in. If you’re having trouble with this, try activating your specialist weapon around a horde - this will force them to respawn and a Stoker will likely take one’s place. Hope this helps! (I did this round 34 on casual and had exactly 3 Stokers.)

Hey guys and girls, just a quick tip for you all, just play on rush mode, it counts and it's so much easier then soloing past round 35 or 4 manning up to round 20 as you have to kill the stoker in the special spot so yeah just play rush mode (easier on solo)

Yep, I also got it this way. I'm not sure whether the entire match of Rush counts as one round or one rush (stuck in a location) counts as a round.

It was easier for us in the classic mode on the easy difficulty level - it’s not difficult to survive the waves.
Somewhere from wave 19 there is a chance for 3 stokers to appear. At wave 25+ there can already be 4-5 out there.
