Ancestors will spawn after certain generators are started, and there will additionally be 3 more at the end of the level when defending the final area.
To get this achievement, make sure you pick up a hypno knife which will randomly spawn in one of the green crates. Now, once an ancestor spawns, use an overcharged shot from an NX-1 Disruptor to get the shields off, and then throw your hypno knife at the ancestor.
You will get a "hit-mark" an the ancestor but the hypno knife will be ineffective, although the achievement will unlock:

Same here, did it on normal and hardcore, probably 10+ times, no achievement

YOU WISH! Hypno Knife a ancestor! Where I show you is 1 of the easiest ways I found. Make sure you shoot the shield off before you throw the knife~! The Ancestor will always appear in the hallway when the 2nd garage generator is initiated! HAPPY HUNTING

For this trophy, you will need a Hypno Knife. You can find a Hypno Knife in grey boxes spread all over the map. When you have found one, all you need to do is aim at the Ancestor and throw it by using the button you use to throw tactical grenades. Ancestors spawn on the second generator in both the Garage and Gas Station area.

First find yourself a Hypno Knife, then just wait for a Ancestor to come to one of the generators and Hypno knife it.

This is simple to do, but finding a Hypno Knife may be a pain. Search the green boxes for a Hypno Knife, then you must hit the ancestor when their shield is down.
