Main Profile (Offense) - Rushing Plays: Cowboy
Secondary Profile (Defense) - 4-3 Base: Arrow
Using this rush play against this defense, I was able to get about 8-12 yards per play. It may not be the best method but it is consistent and does not require you to control your second controller at all. You should be able to just run straight. Do this for about 10 plays to get a total of 100 rush yards to unlock it. You can check your progress in the pause screen stats.

This might have to be 100 yards with 1 player, because I needed to go to ~120 yards, so maybe fumbles recovered by your team won't count for rushing yards for the RB
