In order to unlock Richter Mode, you need to get the good ending by using the Salvation spell on the vampire sisters during their boss fight.
After you finish the game, in order to select Richter mode, you MUST select a new file and you will be able to select Richter mode. Then you simply need to finish all of the painting bosses to fight Dracula. The other boss fights in the main castle are not required.
